County Route 546 is a north–south route within the Town of Brant. Route 546 begins at its south end at the Collins Town Line. Route 546 heads north through the Town of Brant intersecting Mile Strip Rd. (CR 93), then North Collins Rd. (NY 249) before coming to its north end at Cain Rd. (CR 491) which is actually on the Evans-Brant town line.

(Mile Block Road actually begins at Seneca Road in the Town of Collins and within the Cattaraugus Indian Reservation. Since generally there are no county routes within Indian Reservations, Route 546 begins once Mile Block Road leaves the reservation, and at the same time leaves Collins into the Town of Brant.)

Route 578 begins at its southern end at an intersection with Clinton St. (NY 354) and Two Rod Road (unsigned NY 954G and former New York State Route 358) in Marilla. (Two Rod Road continues south from here as NY 954G). Route 547 travels north into the Town of Alden, crossing Cayuga Creek and then intersecting Broadway (US 20) within the town. Route 547 then comes to an end at Westwood Rd. (CR 343)

Route 548 intersects well traveled Baseline Rd. (CR 249) at 2.67 miles (4.30 km) and well-traveled South Parkway (which is NYS Reference Route 957B) at 3.17 miles (5.10 km).

Kenmore Avenue is numbered as three separate county routes along its path: CR 189 in the town of Tonawanda from Grand Island Boulevard (unsigned NY 950C) to the Buffalo city line, CR 550, and CR 307 from NY 265 to Main Street (NY 5) at the Amherst-Buffalo line.

(NY 277 comes from its south end on Boston State Rd. to meet up with Chestnut Ridge Road which continues north from here as NY 277).

County Route 554 known as Youngs Road, this route begins at its south end at Aero Dr. (CR 324) near the southwest–northeast runway for Buffalo-Niagara International Airport. Just after beginning Route 554 enters the Town of Amherst, however the area is known as Williamsville because of its proximity to the village. Youngs Rd. intersects Wehrle Dr. (CR 290), a busy east–west route in Amherst at an 8th of mile. After that intersection, route 554 heads north and is bordered on its east side by Erie Community College (North Campus) all the way to its north end at Main St. (NY 5).

Route 559

County Route 559 the Tonawanda Creek Road portion that lies in Clarence. Route 559 runs east–west beginning at its west end at its intersection with Transit Rd. (NY 78) in an area of Clarence called "Millersport." (Tonawanda Creek Road continues west as CR 2). Route 559 travels east to its end at Rapids Rd. (CR 43) and Salt Rd. (Erie 560 & Former NY 268). (New York State Route 268 formerly was routed on Tonawanda Creek Road from Salt Road, and ended at Transit Road NY 78).^{[citation needed]}

In 2008, work was started on replacing a section of Tonawanda Creek Road in Clarence.^[3] This is because of detours for travelers when they intersect Transit Road. There was a cave-in on CR 559 in June 2004^[3] and has since been repaired as of mid-2009.^{[citation needed]}

County Route 560 is a major thoroughfare through and north of Clarence. The route is 6.67 miles (10.73 kilometres) long, stretching from New York State Route 5 in Clarence to CR 43 and 559 north of Clarence Center. The route is part of the old alignment of New York State Route 268, which included CR 559 as well.

County Route 562 is an east–west route within the Town of Colden. Named Heath Road, Route 562 begins at the intersection of State Rd. (NY 240) and Boston-Colden Rd. (CR 222) at its west end in the hamlet of Colden within the Town of Colden. Route 562 then travels east through Blanchard Hill to its east end at Hayes Hollow Rd. (CR 397). (Heath Road continues east past the intersection with Hayes Hollow another 1.4 miles (2.3 km) to Center St. (CR 33)).

County Route 563 is an east–west route that begins in the Town of Colden and ends up in the Town of Sardinia. Route 563, named Foote Rd. begins at the intersection of Glenwood-East Concord Rd. (NY 240 / CR 30) and Morse Rd. (CR 433). (CR 33 runs concurrent with NY 240) Route 563 comes to its east end in Sardinia at Pratham Rd. (CR 407).

County Route 571 named Center Road, is an east–west roadway in West Seneca. Route 571 begins at its WEST intersection with Seneca St. (CR 215) and travels east to an intersection with busy Union Rd. (NY 277) at one and a half miles, then comes to an end at its EAST intersection with Seneca St. (CR 215), Lein Rd. (CR 154), and Bullis Rd. in an area of town called "East Seneca." Route 571's entire length overlaps New York State Route 16 and is paralleled by the Aurora Expwy. (NY 400).

County Route 572 is a busy north–south route in the Town of Aurora, Town of Elma, and the Village of East Aurora. Route 572 begins at its south end in East Aurora at US 20A (Main St./Hamburg St.). Route 572 travels north as Buffalo Street in East Aurora, Buffalo Road in Aurora, and Seneca Street in Elma. Route 572 in its entireity overlaps the NY 16/NY 78 concurrency from US 20 to US 20A. Intersections along route 572 include many local roadways including Jamison Road (CR 574 & Former NY 422). Route 572's north end is at Transit Rd. (US 20) and CR 215. New York State Route 78 ends its concurrency with NY 16 and heads north on Transit Road with US 20, while New York State Route 16 continues on Seneca Street (also as CR 215) in West Seneca towards Buffalo.

County Route 573 is an east–west route in the Village of Akron and Town of Newstead. Route 573 begins at its western end at Cedar St. (NY 93) in Akron and is named John St. Route 573 travels east and turns northeast onto Bloomingdale Rd. and travels into the Town of Newstead. Route 573 then comes to an end at the Genesee County Line. (Route 573 used to be New York State Route 267, which ran from Akron to Basom (a hamlet in Alabama, New York). However NY 267 was decommissioned on August 14, 1980 and replaced by the Route 573 designation in Erie County).

County Route 574 is a major thoroughfare north of East Aurora. Known locally as Jamison Road, the road supplies the hamlet of East Elma. The road starts at Seneca St.(NY 16/NY 78/CR 572) south of the hamlet Spring Brook and ends at Two Rod Road (unsigned NY 954G) in the Town of Marilla. This county route was formerly New York State Route 422, but was decertified on September 25, 1980 and was given to the county.

(CR 575).

Big Tree Road continues east past Bayview Rd. (CR 575) as just Bayview Road and begins as US 20A at Southwestern Blvd. (US 20) in Hamburg.

Route 580 begins at the City of Tonawanda - Town of Town of Tonawanda line (at its west end) and travels to Niagara Falls Blvd. (US 62) its east end. Niagara Falls Blvd. divides the Town of Tonawanda and Town of Amherst. There is an intersection with Colvin Blvd. (CR 303) within the town. (Ellicott Creek Road continues west into the City of Tonawanda and ends at Young Street.)
