The following is a list of county routes in Erie County, New York from Routes 33 to 64. Most of the routes act as primary roads in the less developed areas and also serve to interconnect the various villages and hamlets of the county. Municipalities served include the cities of Buffalo and Tonawanda, and the villages of Angola, Evans, Farnham, Lancaster, Springville, East Aurora and Depew.

County Route 33 Location: Colden, Aurora Length: 10.41 mi[1][2] (16.75 km) County Route 33 is one of the longest county roads in Erie County. It stretches for 10.3 miles (16.6 km) from CR 408 in Colden to the triplex of US 20A/NY 16/NY 78 in East Aurora. The road goes north to south and crosses through Colden, Aurora and East Aurora.
Route 33 begins at an intersection with Erie CR 408 (Crump Road) in Colden.[3] The road goes north, intersecting with Erie CR 240 (Holland-Greenwood Road) in Colden. The two roads are concurrent for a short distance, with Route 408 continuing to the east afterwards. CR 396 intersects at 3.0 miles (4.82 kilometers) and continues north. CR 85 (Blakeley Corner Road) intersects at 8.3 miles (13.35 kilometres) in Aurora. CR 112 comes afterward, intersecting just outside of East Aurora. Route 33 comes to an end at Routes 16, 78 and 20A in downtown East Aurora.

County Route 34 Location: {{{location}}} Length: 1.40 mi[2] (2.25 km) County Route 34 is a short thoroughfare in Irving and Farnham. Route 34 is 1.4 miles (2.3 km) long, stretching from CR 111 in Irving to the intersection with NY 5 and NY 249 in Farnham. There are no intersections in between and is known as Lotus Point Road.[4]

County Route 37 Location: {{{location}}} Length: 5.57 mi[1] (8.96 km) County Route 37 is a relatively minor thoroughfare in the town of Clarence. The road is 5.57 miles (8.96 km), stretching from Route 78 and CR 192 (Maple Road) in Amherst to CR 560 (Salt Road) in Clarence. The road is also an access to Brookside Country Club in Clarence.
Route 37 begins at an intersection with CR 192 and NY 78 (Transit Road) in Amherst.[5] Route 37 passes some local roads, intersecting with CR 276 (Harris Hill Road) at .9 of a mile. Route 37 passes to the north of Brookside Country Club and intersects with CR 216 (Goodrich Road) at 3 miles (4.8 km). Route 37 makes a curve to the north, and then back to the east. Route 37 intersects with CR 269 (Strickler Road) at 4.5 miles (7.2 km). Route 37 passes to the north of downtown Clarence and comes to an end at CR 560 (Salt Road).

County Route 39 Location: {{{location}}} Length: 3.02 mi[1] (4.86 km) County Route 39 is the only county-maintained road that accesses downtown Langford. Route 39 is 3.02 miles (4.86 km) long and stretches from New York State Route 249 and New York State Route 75 in Langford to the Concord city line, where it becomes County Route 50. The road is known as Langford-New Oregon Road throughout its whole distance.
Erie CR 39 begins at an intersection with Routes 75 and 249 in downtown Langford.[6] Route 39 continues east, intersecting with local roads. Route 39 intersects with CR 448 (New Oregon Road) and comes to an end at CR 50 in Concord.

County Route 40 Location: {{{location}}} Length: 1.35 mi[1] (2.17 km) County Route 40 is a short thoroughfare in the town of Collins.[7] The road is .90 of a mile long and connects County Route 508 (Becker Road) to New York State Route 39 in Collins Center. The road heads north to south and is known as Collins Center – Zoar Road.

County Route 41 Location: {{{location}}} Length: 8.38 mi[1] (13.49 km) County Route 41 is a north–south route named Versailles Plank Rd., and travels through the towns of Brant, Evans and just crosses into Eden near its northern end. Route 41 begins at New York State Route 249 (North Collins Rd.) in the Town of Brant. (Versailles Plank Road does continue south of NY 249 in Brant). Route 41 crosses into Evans at an intersection with Cain Rd. (CR 491) at 1-mile (1.6 km). Route 41 continues north through Evans, but as a result of Interstate 90 (NYS Thruway), Versailles Plank is broke in two sections, both connected to CR 32 named Evans Center Road at 4.2 miles (6.8 km) north from its south end. Route 41 continues west of I-90 and continues north intersecting US 20 (Southwestern Blvd.) at 5.1 miles (8.2 km), Sturgeon Point Road (CR 18) at 6.4 miles (10.3 km), and ends at South Creek Rd. (CR 476) at 8.38 miles (13.49 km) in the hamlet of North Evans.

County Route 42 Location: {{{location}}} Length: 5.35 mi[1] (8.61 km) County Route 42 is generally an east–west route in the Town of Newstead and a short distance in Clarence. Route 42 begins its west end at the Erie - Niagara County Line, and intersects CR 43 (Rapids Road's continuation East) just into Erie County. Route 42 heads east out of Clarence and into Newstead, coming to its eastern terminus at Maple Rd. (NY 93).

(CR 44), and Clark St. (CR 51) in a hamlet of Hamburg named Armor. Route 44 heads east through Orchard Park and at 0.4 miles (0.64 km), Powers Rd. (CR 370) splits off and heads east as Route 44 continues northeast (east) until Route 44's east end at the intersection of Duerr Rd. (CR 463) and Armor-Duells continuation as New York State Route 952J, an unsigned reference route. (Three-tenths of a mile later Armor Duells Rd. (NY 952J) has access to US 219).

The road lies entirely in Amherst. Route 45 begins at an intersection with Erie County Route 301 (Sweet Home Rd.) at its west end. Route 45's east end is at an intersection with NY 78 (Transit Rd.) at the Amherst-Clarence town line.

Route 47, named Mill Road/Mill Street, runs from Davis Rd. (NY 240) in the hamlet of Jewettville in the southwest part of Aurora, east, then northeast towards East Aurora. Route 47 comes to an end at US 20A (Quaker St./Hamburg St.) within the village. Mill Road has an intersection with Blakeley Corners Rd. (CR 85) at 1.8 miles (2.9 km).

County Route 49 Location: {{{location}}} Length: 6.4 mi[2] (10.30 km) County Route 49 is an east–west route in the Towns of Concord named Genesee Road. Route 49 begins at its west end at the its intersection with Wyandale Rd. (CR 50) /Mortons Corners-Wyandale Rd. (CR 106) and Genesee's continuation as CR 73. Route 49 then heads east and intersects US 219 (Southern Expressway) at 2.7 miles (4.3 km), and CR 228 (Springville-Boston Rd.) at 4.4 miles (7.1 km). Route 49 comes to its east end at an intersection in the hamlet of East Concord with NY 240/CR 198, CR 27, CR 432, and Genesee's continuation as CR 409.
Genesee Road itself is 32 miles (51 km) long, and stretches from Quaker St. (CR 498) in North Collins to New York State Route 39 in the Town of Eagle in Wyoming County. Genesee Road consists of three county routes: CR 73 (7.7 miles or 12.4 kilometres) from Quaker Street (CR 498) in North Collins to the intersection of Wyandale Road (CR 50) and Mortons Corners – Wyandale Road (CR 106), CR 49 from there to New York State Route 240 in Concord, and CR 409 (8.5 miles or 13.7 kilometres) from NY 240 to the Wyoming County line.
