County Route 519 is a short east–west route the extreme north part of the Town of Concord, located in the hamlet of Fowlerville, just south of the Town of Boston Line. Route 519 runs from CR 420 (Trevett Rd.) east to CR 228 (Springville-Boston Rd.), the old routing of US 219 prior to the construction of the Southern Expressway.

Route 521

County Route 521 is an east–west route the northeastern part of the Town of Tonawanda named Creekside Drive. Route 521 begins its western end at the Tonawanda City Line. (West into the city the roadway is named East Niagara Street) Creekside Drive travels along the south side of Tonawanda Creek/Erie Canal for its entire length, and also passes through part of Ellicott Creek Park[1] on its way to its eastern terminus. Route 521 comes to an end at Niagara Falls Blvd. (US 62). Creekside Drive itself continues east into the Town of Amherst as a locally maintained roadway.

County Route 523 is a busy east–west route through the Town and Village of Lancaster and Cheektowaga named Como Park Boulevard. Route 523 begins its western end at Union Road (NY 277), a major north–south thoughofare in Cheektowaga. Route 523 travels east through south-central Cheektowaga, south of the Village of Depew, and intersects Transit Road (US 20/NY 78) at the Lancaster Town Line. Como Park Blvd. continues east on into the Village of Lancaster at 3.6 miles (5.8 km). After intersecting local village roadways, Route 523 comes to an end at Como Lake Park within the Village of Lancaster.

County Route 524 is a north–south route the Town of Tonawanda named Two Mile Creek Road, named after a nearby creek of the same name. Route 524 begins its southern end at CR 189 (Kenmore Ave.) near CR 189's intersection with NY 325. 524 heads north, paralleling I-190 for a short distance before coming to an intersection with Grand Island Blvd. (NY 324) at 0.4 miles (0.64 km). The route continues north towards the City of Tonawanda, passing on the west side of Sheridan Park and crossing over I-290 at 1.3 miles (2.1 km), and now closely paralleling Two Mile Creek. Route 524 ends at 1.6 miles (2.6 km) at the Tonawanda City Line where the county route designation ends. Two Mile Creek Road continues north on the western edge of the City to the roadway's north end at Niagara Street (NY 266).

County Route 528 is an east–west route the Town of Elma named West Blood Road. Route 528 begins its western end at Seneca St. (NY 16/NY 78/CR 572) 1.5 miles northeast of East Aurora. Route 528 travels through the south part of Elma, north of the Town of Aurora, and village of East Aurora. At 1.6 miles (2.6 km) there is an intersection with Maple Street which can take you into East Aurora to the south or to NY 400 (Aurora Expwy.) to the north. West Blood Road/CR 528 is 3.9 miles (6.3 km) long and ends at East Aurora-Porterville Road (CR 6) at the Elma-Marilla Town Line, west of Porterville.

Route 534

County Route 534 is a 1.8-mile (2.9 km) east–west route the western part of the Town of West Seneca. Route 534 begins its west end at the Buffalo City Line. Mineral Springs Road itself actually begins at Seneca St. (NY 16), 0.7 miles (1.1 km) to the west, however since there are no county routes within city limits, route 534 begins at the Buffalo city line with West Seneca. From there 534 heads east passing under Interstate 90 (New York State Thruway) and shortly after intersecting Harlem Rd. (New York State Route 240). After that intersection Mineral Springs Road/Route 534 head in a southeasterly direction to an east end at Indian Church Rd. (CR 109). CR 534 roughly parallels Buffalo Creek which lies north of the roadway in varying distances.

Route 542 continues east, intersecting a few local roadways and passes the Erie County Society for the Prevention of Cruelty to Animals, and comes to an end at New York State Route 265 (Military Rd.)

Route 543

County Route 543 is a general southwest–northeast route located in the south part of the Town of Boston and extreme north part of the Town of Concord. Route 543 begins at CR 436 (Brown Hill Rd.) and heads north entering Boston while paralleling US 219 (Southern Expwy.) which lies on the west side. At eight tenths of a mile, Pfarner Rd/Route 543 turn northeast. Prior to the construction of the Southern Expressway, Pfarner Road connected to Zimmerman Rd. (CR 437) a half mile on the other side of the expressway. As Route 543 heads northeast it ends briefly at West Tillen Rd (CR 233) at 1.7 miles (2.7 km). However, Route 543 once again continues three tenths of a mile east of there at Mill Street, which route 543 continues as into the hamlet of Boston. At 2.2 miles (3.5 km) Route 543 ends at an intersection with CR 227 (Boston State Rd.) and CR 443 (Boston Cross Rd.).
