The southern terminus of the route is at the Delaware state line in New Garden Township, where it continues as Delaware Route 41. The northern terminus is at U.S. Route 30 just north of Gap.
PA-41, also known as Gap-Newport Pike (or Newport-Lancaster Pike), serves as the primary thoroughfare between Lancaster's eastern suburbs and Wilmington, Delaware; as the original southern terminus of PA / DE 41 was in Newport, the highway's nickname persists. Frequent accidents have motivated PennDOT's many improvements along its length, as well as the addition of signage reminding motorists to drive safely.

Route description
Lancaster County
South-bound PA 41 begins at US 30 in Gap; less than a mile on, its first minor junction is the eastern terminus of PA 741.
Chester County
In Londonderry Township, PA 41 meets with northern terminus of PA 796 and then intersects PA 926 immediately afterwards. The highway then intersects with PA 10 in Cochranville. London Grove Township houses PA-41's major junction with US 1 at an interchange one mile southeast of PA 841 near Chatham. In Avondale, PA 41 meets with Baltimore Pike a major road in Chester & Delaware Counties. Just before the state line, it has a mini-interchange with SR 3013 (Limestone Road) at Kaolin, which eventually turns into DE 7 upon crossing the border. The southern terminus of PA 41 is in Kennett Township, on the state line (Delaware), where it continues as DE 41 towards the city of Wilmington.
