Its western terminus is at Pennsylvania Route 74 in Lower Chanceford Township north of Delta and west of Holtwood, and its eastern terminus is at Pennsylvania Route 82 in Coatesville.

Route description
York and Lancaster Counties
Pennsylvania Route 372 heads to the northeast of Pennsylvania Route 74 on Holtwood Road. It then crosses the Susquehanna River on the Norman Wood Bridge just south of the Holtwood Dam into Martic Township in Lancaster County. It heads to the northeast and enters the northern part of Drumore Township, running just south of the border with Providence Township. The route intersects Pennsylvania Route 272 in the village of Buck, where it crosses into East Drumore Township and the name changes to Buck Road. PA 372 then heads east and enters the borough of Quarryville, where it becomes State Street. It intersects U.S. Route 222 in the western part of town and the northern terminus of Pennsylvania Route 472 in the eastern part of the town. PA 372 then leaves Quarryville and enters Eden Township, where it becomes Valley Road. It continues into Bart Township, where it meets Pennsylvania Route 896. PA 372 then turns north for a concurrency with PA 896 on Georgetown Road. It splits from PA 896 in the village of Georgetown by heading east on Christiana Pike. It crosses into Sadsbury Township and then into the borough of Christiana, where it becomes Water Street. Within Christiana, PA 372 turns south onto South Bridge Street before turning east on Valley Avenue and crossing the East Branch of the Octoraro Creek into Chester County.
Chester County
Pennsylvania Route 372 briefly passes through West Sadsbury Township before entering the borough of Atglen. In Atglen, PA 372 turns south onto Main Street before turning east on Lower Valley Road and intersecting Pennsylvania Route 41. It then re-enters West Sadsbury Township and proceeds east to the borough of Parkesburg. In Parkesburg, PA 372 turns north onto South Limestone Road and then bends east onto First Avenue. It intersects Pennsylvania Route 10 in the eastern part of the borough. PA 372 then leaves Parkesburg and heads east on Valley Road, passing through Sadsbury and Valley Townships. It then enters the city of Coatesville where it ends at an intersection with Pennsylvania Route 82 in the southwestern corner of the city.
