Methotrexate . This is afolic acid analogue, and owing to structural similarity with it binds and inhibits the enzymedihydrofolate reductase , and thus prevents the formation oftetrahydrofolate . Tetrahydrofolate essential forpurine andpyrimidine synthesis, and this leads to inhibited production ofDNA ,RNA and proteins (as tetrahydrofolate is also involved in the synthesis of amino acidsserine andmethionine ).Other examples include
trimethoprim ,pyrimethamine andpemetrexed .Since, antifolates interfere with metabolism (of
nucleotide s), they are categorized asantimetabolite s.
