Mir-148/mir-152 microRNA precursor family

Mir-148/mir-152 microRNA precursor family

In molecular biology, miR-148 is a microRNA whose expression has been demonstrated in human ( [http://microrna.sanger.ac.uk/cgi-bin/sequences/mirna_entry.pl?acc=MI0000253 MI0000253] ), mouse ( [http://microrna.sanger.ac.uk/cgi-bin/sequences/mirna_entry.pl?acc=MI0000550 MI0000550] ), rat ( [http://microrna.sanger.ac.uk/cgi-bin/sequences/mirna_entry.pl?acc=MI0000616 MI0000616] ) and zebrafish ( [http://microrna.sanger.ac.uk/cgi-bin/sequences/mirna_entry.pl?acc=MI0002015 MI0002015] ).cite journal | last = Lagos-Quintana | first = M | coauthors = Rauhut R, Yalcin A, Meyer J, Lendeckel W, Tuschl T | year = 2002 | title = Identification of tissue-specific microRNAs from mouse | journal = Curr Biol | volume = 12 | pages = 735–739 | pmid = 12007417 | doi = 10.1016/S0960-9822(02)00809-6] [cite journal | last = Kim | first = J | coauthors = Krichevsky A, Grad Y, Hayes GD, Kosik KS, Church GM, Ruvkun G | year = 2004 | title = Identification of many microRNAs that copurify with polyribosomes in mammalian neurons | journal = Proc Natl Acad Sci U S A | volume = 101 | pages = 360–365 | pmid = 14691248 | doi = 10.1073/pnas.2333854100] [cite journal | last = Chen | first = PY | coauthors = Manninga H, Slanchev K, Chien M, Russo JJ, Ju J, Sheridan R, John B, Marks DS, Gaidatzis D, Sander C, Zavolan M, Tuschl T | year = 2005 | title = The developmental miRNA profiles of zebrafish as determined by small RNA cloning | journal = Genes Dev. | volume = 19 | pages = 1288–1293 | pmid = 15937218 | doi = 10.1101/gad.1310605] miR-148 has also been predicted in chicken ( [http://microrna.sanger.ac.uk/cgi-bin/sequences/mirna_entry.pl?acc=MI0001189 MI0001189] ).

These predicted hairpin precursor sequence are related to those of miR-152, which has been expressed in mouse ( [http://microrna.sanger.ac.uk/cgi-bin/sequences/mirna_entry.pl?acc=MI0000174 MI0000174] ) and is predicted in human ( [http://microrna.sanger.ac.uk/cgi-bin/sequences/mirna_entry.pl?acc=MI0000462 MI0000462] ).

The hairpin precursors (represented here) are predicted based on base pairing and cross-species conservation; their extents are not known. In this case, the mature sequence is excised from the 3' arm of the hairpin.

Targets of miR-148

MicroRNAs act by lowering the expression of genes by binding to target sites in the 3' UTR of the mRNAs. However recently it was shown by Duursma and colleagues that miR-148 down regulates Dnmt3b by binding to a region in the protein coding region.cite journal |author=Duursma AM, Kedde M, Schrier M, le Sage C, Agami R |title=miR-148 targets human DNMT3b protein coding region |journal=RNA |volume=14 |issue=5 |pages=872–7 |year=2008 |month=May |pmid=18367714 |doi=10.1261/rna.972008 |url=]
