She was a daughter of Frederick Francis I, Grand Duke of Mecklenburg-Schwerin and Princess Louise of Saxe-Gotha-Altenburg.

Marriage
She married Danish heir apparent Prince Christian of Denmark and Norway in 1806. In 1808, She gave birth to their only surviving son and later King of Denmark Frederick VII of Denmark.
Duchess Charlotte Frederika allegedly had an affair with the French singer and composer Edouard Du Poy. For this reason, her husband divorced her in 1810, sent her into internal exile in Horsens, and prohibited her to see her son again.
Later life
She spent the next years of her life in a palace in Horsens, in Jutland, where she continued different affairs with local officers. In 1830, Charlotte Frederica went to Italy, finally settling in Rome. She later converted to the Catholic faith. Charlotte Frederica died on 1840.
