He was the second son of Friedrich I von Mecklenburg-Schwerin (1638-1688) and his wife, née Christine von Hessen-Homburg (1653–1722), and succeeded his brother Friedrich Wilhelm von Mecklenburg-Schwerin as duke in 1713.
Karl Leopold married three times. His first wife was Sophia Hedwig of Nassau-Dietz (1690–1734), daughter of Heinrich Kasimir II von Nassau-Dietz (1657–1696) and his wife, née Henriëtte Amalia von Anhalt-Dessau (1666–1726). They married on 27 May 1709 in Leeuwarden and divorced in 1710. There were no children of this marriage.
His second wife was Christine von Lepel (1692–1728), daughter of Nicolaus Friedrich von Lepel and his wife née Leveka von Plessen. They married 7 June 1710 at Doberau, and divorced on 2 October 1711. There were no children of this marriage.
His third wife was Grand Duchess Jekaterina Ivanovna of Russia (1692–1733), daughter of Tsar Ivan V of Russia (1666–1696) and his wife, née Praskovaya Feodorovna Saltykova. They married on 19 April 1716 at Danzig. There was one daughter born of this marriage, Elisabeth Katharina Christina of Mecklenburg-Schwerin, known as Anna Leopoldovna. She married Anton Ulrich, Duke of Brunswick-Bevern-Luneburg, and had 5 children. She served as Regent of Russia for her son Ivan VI from 1740-1741.
