Wayson stain

Wayson stain

The Wayson stain is a modified methylene blue stain, originally used for diagnosing Bubonic plague. [cite journal|author=Meyer KF and Batchelder AP|year=1926|title=A disease in wild rats caused by "Pasteurella muricida," n. sp.|journal=J Infect Dis|volume=39|pages=386–412] With this stain, "Yersinia pestis" appears purple with a characteristic safety-pin appearance, [cite journal|author=Inglesby TV, Dennis DT, Henderson DA, "et al."|title=Plague as a biological weapon|journal=JAMA|year=2000|volume=283|pages=2281–2290] which is due to the presence of a central vacuole. It is used along with the Giemsa and Wright's stains to rapidly detect potential biowarfare attacks [ [http://www.medscape.com/content/2001/00/42/14/421420/421420_tab.html Medscape.com Clues to the Specific Etiologic Agent Used in a Biological Attack] ]

It has also been investigated as a possible cheaper and faster way to detect Melioidosis. [cite journal|author=Sheridan, Elizabeth A., Ramsay , Andrew R., Short, Jennifer M., Stepniewska , Kasia, and Wuthiekanun, Vanaporn, Simpson , Andrew J. H. Simpson|title= [http://www.pubmedcentral.nih.gov/articlerender.fcgi?artid=1865910 Evaluation of the Wayson Stain for the Rapid Diagnosis of Melioidosis] |journal=Journal of Clinical Microbiology|year=2007|volume=45(5)|pages=1669–1670]

It is a useful alternative to the Gram or Loeffler’s Methylene Blue stains, especially for detecting Yersinia Entercoliteiea which is often found in spoiled food [ [http://www.clpmag.com/issues/articles/2001-08_22.asp CLP Magazine Aug 2001] ]
