The A749 road connects East Kilbride New Town with Glasgow City Centre. The road starts off at a roundabout called "The Whirlies", with a junction for the A725 road, starting as a dual carriageway. The road continues and it meets another roundabout for the A749 spur which leads onto the A725 road. The road continues and goes past the Kingsgate Retail Park. After a traffic light junction the road becomes National Speed Limit through open countryside descending.

Around two miles north-west there is a grade separated junction for the B759 road to Cambuslang and Carmunock and the road reduces to a 40 mile per hour speed limit. The road continues after another roundabout where an unclassified road for Cathkin joins. The road continues for another brief spell as a 40 mile per hour dual carriageway.

Soon afterwards the road drops to a 30 mile per hour zone and the dual carriageway section ends. Passing through the Burnside the road becomes urban and continues this down to Rutherglen. It crosses the River Clyde and enters Dalmarnock still heading in a northwesterly direction.

The road approaches Bridgeton and it continues past the B763 road as Dalmarnock Road, although road signs suggest Dunn Street heading west is the A749 when this is incorrectFact|date=August 2008|require reliable source for signage issues. At Bridgeton the road switches to London Road and carries on west to Glasgow City Centre passing the Barras flea market. It forms a brief part of the one way system along with the A89 road on the eastern edge of the city of Glasgow.

It ends at a grade junction with the A8 road as Trongate continues into the heart of Glasgow City Centre unclassified.
