The 27th Regiment Illinois Volunteer Infantry was an infantry regiment that served in the Union Army during the American Civil War.

The 27th Illinois Infantry was organized at Camp Butler, Illinois and mustered into Federal service on August 10, 1861.
The regiment was mustered out on September 20, 1864 at Springfield, Illinois. The veterans and recruits from this regiment were transferred to the 9th Illinois Volunteer Infantry Regiment.
Total strength and casualties
The regiment suffered 7 officers and 96 enlistedmen who were killed in action or who died of their wounds and 2 officer and 83 enlistedmen who died of disease, for a total of 188 fatalities.
