LYK5

LYK5

Protein kinase LYK5, also known as LYK5 or STRAD, is a human protein and also denotes the gene encoding it.cite web | title = Entrez Gene: LYK5 protein kinase LYK5| url = http://www.ncbi.nlm.nih.gov/sites/entrez?Db=gene&Cmd=ShowDetailView&TermToSearch=92335| accessdate = ] cite journal | author = Baas AF, Boudeau J, Sapkota GP, Smit L, Medema R, Morrice NA, Alessi DR, Clevers HC | title = Activation of the tumour suppressor kinase LKB1 by the STE20-like pseudokinase STRAD | journal = EMBO J. | volume = 22 | issue = 12 | pages = 3062–72 | year = 2003 | month = June | pmid = 12805220 | pmc = 162144 | doi = 10.1093/emboj/cdg292 | url = | issn = ]

Function

Endogenous LKB1 and LYK5 form a complex in which LYK5 activates LKB1, resulting in phosphorylation of both partners. Removal of endogenous LYK5 by small interfering RNA abrogates LKB1-induced G1 phase arrest.

Disease linkage

Mutations in the LYK5 gene are associated with polyhydramnios, megalencephaly and symptomatic epilepsy (collectively known as the PMSE syndrome).cite journal | author = Puffenberger EG, Strauss KA, Ramsey KE, Craig DW, Stephan DA, Robinson DL, Hendrickson CL, Gottlieb S, Ramsay DA, Siu VM, Heuer GG, Crino PB, Morton DH | title = Polyhydramnios, megalencephaly and symptomatic epilepsy caused by a homozygous 7-kilobase deletion in LYK5 | journal = Brain | volume = 130 | issue = Pt 7 | pages = 1929–41 | year = 2007 | month = July | pmid = 17522105 | doi = 10.1093/brain/awm100 | url = | issn = ]
