Acetylcholinesterase, also known as AChE, is an
enzyme that degrades (through itshydrolytic activity) the neurotransmitteracetylcholine , producingcholine and anacetate group. It is mainly found atneuromuscular junction s andcholinergic synapse s in thecentral nervous system , where its activity serves to terminatesynaptic transmission . AChE has a very high catalytic activity - each molecule of AChE degrades about 5000 molecules of acetylcholine per second. The choline produced by the action of AChE is recycled - it is transported, throughreuptake , back intonerve terminal s where it is used to synthesize new acetylcholine molecules.cite book | author = Purves, Dale, George J. Acetylcholinesterase is encoded by the single AChE gene; and the structural diversity in the gene products arises from alternative mRNA splicing and
post-translational associations of catalytic and structural subunits. The major form of acetylcholinesterase found in brain, muscle, and other tissues is the hydrophilic species, which forms disulfide-linked oligomers with collagenous, orlipid -containing structural subunits. The other, alternatively-spliced form, expressed primarily in the erythroid tissues, differs at theC-terminus , and contains a cleavablehydrophobic peptide with a GPI-anchor site. It associates with membranes through thephosphoinositide (PI) moieties added post-translationally. Acetylcholinesterase is the target of many
nerve gas es, particularly theorganophosphates (e.g.Sarin ). These agents - known ascholinesterase inhibitor s - block the function of acetylcholinesterase and thus cause excessive acetylcholine to accumulate in thesynaptic cleft . The excess acetylcholine causes neuromuscularparalysis (i.e. interminablemuscle contraction s) throughout the entire body, leading to death byasphyxiation .ee also
