It is the 5th installment of the series, which began with NCAA College Basketball 2K3. It features former Duke and current Orlando Magic guard J.J. Redick on the cover. It is produced by 2K Sports. It was the first college basketball game for the Playstation 3. Redick was a special correspondant to the development of the game and added his signature shot style in motion capture.

Trivia
- In Legacy Mode if the player views the messages there will be some spam on the list of unread messages advertising useless products.
- On the Selection Sunday Board the top or bottom half of the teams on the Bubble will get in to the NCAA Tournament.
- Coaches Bob Knight, Mike Krzyzewski, Jim Boeheim, and Rick Pitino do not lend their likenesses to the game but coaches Roy Williams, Lute Olson, Billy Donovan, John Calipari, Jay Wright, and Sidney Lowe do.
- There is a preview show with hosts Greg Gumbel and Clark Kellogg.
- The player has an option to give fake names to players on all of the teams at the start of the game.
- If you play as the Connecticut Huskies, they are called Cincinnati in the Xbox 360 version of the game.
