LocusSupplementaryData =Luciferase is a generic name for
enzyme s commonly used in nature forbioluminescence . The most famous one isfirefly luciferase (EC number|1.13.12.7) from thefirefly "Photinus pyralis". In luminescent reactions, light is produced by theoxidation of aluciferin (a pigment), sometimes involvingadenosine triphosphate (ATP). The rates of this reaction between luciferin andoxygen are extremely slow until they arecatalyzed by luciferase, often mediated by the presence ofcalcium ions (an analog ofmuscle contraction ).cite web | url = http://www.chem.qmul.ac.uk/iubmb/enzyme/EC1/13/12/5.html | title = EC 1.13.12.5 | author = | authorlink = | coauthors = | date = | format = | work = IUBMB Enzyme Nomenclature | publisher = Nomenclature Committee of the International Union of Biochemistry and Molecular Biology (NC-IUBMB) | pages = | language = | archiveurl = | archivedate = | quote = | accessdate = 2008-10-02] "Renilla" luciferin reaction is triggered by calcium ions. (citedSeptember 6 ,2006 )] The reaction takes place in two steps:
luciferin + ATP → luciferyl adenylate + PPi
luciferyl adenylate + O2 → oxyluciferin + AMP + light
The reaction is very energy efficient: nearly all of the energy input into the reaction is transformed into light. As a comparison, the incandescent light bulb loses about 90% of its
energy toheat . [General Electric TP-110, page 23, table.]Luciferin and luciferase are not specific molecules. They are generic terms for a substrate and its associated
enzyme (orprotein ) thatcatalyze a light-producing reaction. A variety of organisms regulate their light production using different luciferases in a variety of light-emitting reactions. The most famous are the fireflies, although the enzyme exists in organisms as different as the Jack-O-Lantern mushroom "(Omphalotus olearius )" and many marine creatures. In fireflies, the oxygen required is supplied through a tube in the abdomen called theabdominal trachea . The luciferases of fireflies - of which there are over 2000species - and of theElateroidea (fireflies, click beetles and relatives) in general - are diverse enough to be useful inmolecular phylogeny . The most thoroughly studied luciferase is that of thePhotinini firefly "Photinus pyralis", which has an optimum pH of 7.8.cite journal | author = Steghens JP, Min KL, Bernengo JC | title = Firefly luciferase has two nucleotide binding sites: effect of nucleoside monophosphate and CoA on the light-emission spectra | journal = Biochem. Luciferase can be produced in the lab through
genetic engineering for a number of purposes. Luciferasegene s can be synthesized and inserted into organisms or transfected into cells. Mice, silkworm s, andpotato es are just a few organisms that have already been engineered to produce the protein. Fact|date=September 2008In the luciferase reaction, light is emitted when luciferase acts on the appropriate luciferin substrate. Photon emission can be detected by light sensitive apparatus such as a
luminometer or modified optical microscopes. This allows observation of biological processes.Promega Corporation. [http://www.promega.com/multimedia/bioLum01.htm Introduction to Bioluminescent Assays] . (Animation, requires Flash player)]In biological research, luciferase commonly is used as a reporter to assess the transcriptional activity in cells that are transfected with a genetic construct containing the luciferase gene under the control of a
promoter of interest.cite journal | author = Fan F, Wood KV | title = Bioluminescent assays for high-throughput screening | journal = Assay Drug Dev Technol | volume = 5 | issue = 1 | pages = 127–36 | year = 2007 | month = February | pmid = 17355205 | doi = 10.1089/adt.2006.053 | url = | issn = ] Luciferase can also be used detect the level of cellular ATP in cell viability assays or for kinase activity assays.cite journal | author = Meisenheimer PL, O’Brien MA, Cali JJ | title = Luminogenic enzyme substrates: The basis for a new paradigm in assay design. | journal = Promega Notes | volume = 100 | issue = | pages = 22–26 | year = 2008 | month = September | pmid = | url = http://www.promega.com/pnotes/100/16620_22/16620_22.pdf | issn = ] Additionally proluminescent molecules that are converted to luciferin upon activity of a particular enzyme can be used to detect enzyme activity in coupled or two-step luciferase assays. Such substrates have been used to detect caspase activity and cytochrome P450 activity, among others.Whole animal imaging (referred to as in vivo or, occasionally, ex vivo imaging) is a powerful technique for studying cell populations in live animals, such as mice. Different types of cells (e.g. bone marrow stem cells, T-cells) can be engineered to express a luciferase allowing their non-invasive visualization inside a live animal using a sensitive
CCD camera .This technique has been used to follow tumorigenesis and response of tumors to treatment in animal models.cite journal | author = Lyons SK, Meuwissen R, Krimpenfort P, Berns A | title = The generation of a conditional reporter that enables bioluminescence imaging of Cre/loxP-dependent tumorigenesis in mice | journal = Cancer Res. | volume = 63 | issue = 21 | pages = 7042–6 | year = 2003 | month = November | pmid = 14612492 | url = http://cancerres.aacrjournals.org/cgi/content/abstract/63/21/7042 | issn = ] cite journal | author = Becher OJ, Holland EC | title = Genetically engineered models have advantages over xenografts for preclinical studies | journal = Cancer Res. | volume = 66 | issue = 7 | pages = 3355–8, discussion 3358–9 | year = 2006 | month = April | pmid = 16585152 | doi = 10.1158/0008-5472.CAN-05-3827 | url = | issn = ] Luciferase can be used inblood bank s to determine ifred blood cell s are starting to break down.Forensic investigators can use a dilute solution containing the enzyme to uncover traces of blood remaining on surfaces at a crime scene. Luciferase is a heat sensitive protein that is used in studies onprotein denaturation , testing the protective capacities ofheat shock proteins . The opportunities for using luciferase continue to expand. Fact|date=September 2008ee also
