- Marsa and Lija Athletic were promoted from the First Division to the Premier League.They were replaced with Rabat Ajax and Xghajra Tornadoes,both relegated from Maltese Premier League 2000-01.
- Tarxien Rainbows and Zurrieq were relegated to the Maltese Second Division 2001-02.They were replaced with Mqabba,champions of Second Division 00-01 and Balzan Youths,runners-up.
