The route starts at the Victoria St terminus (at the intersection with Church St, stop 64), then runs along Church St, Richmond, crossing Bridge Rd (stop 61) and Swan St (stop 57). It continues over the Yarra River (into the suburb of South Yarra) along Chapel St, crossing Toorak Rd at stop 49. Continuing down Chapel St, Route 78 terminates at Brighton Rd, Balaclava (stop 36). It is one of the few Melbourne tram lines that does not pass through the CBD.
This service is predominantly run by W class trams with A and Z class trams supplementing stock requirements as needed. The other W exclusive route in Melbourne is the route 30 to Docklands and the City Circle Tram.
